Whether the anabolic effect of insulin-like growth factor-I (IGF-I) in osteoblastic MC3T3-E1 cells is modulated by zinc, an activator of bone formation, was investigated in vitro. After subculture for 3 days, the cells were cultured for 72 h with IGF-I (10(-8) M). The peptide produced a significant increase of protein concentration, deoxyribonucleic acid (DNA) content, and cell number in the cells. <b>These increases were markedly enhanced by the presence of zinc sulfate (10(-5) M), but not zinc-chelating dipeptide (beta-alanyl-L-histidinato zinc; 10(-5) M).</b> Also, the cellular alkaline phosphatase activity was synergistically increased by the presence of both IGF-I and zinc sulfate. Thus, effect was not seen in the presence of both insulin (10(-8) M) and zinc sulfate (10(-5) M). The effect of zinc sulfate to enhance the IGF-I-increased alkaline phosphatase activity and protein concentration in the cells was clearly prevented by the presence of cycloheximide (10(-6) M), staurosporin (10(-8) M), or okadaic acid (10(-7) M) with an effective concentration. However, staurosporin had a partial inhibiting effect on the IGF-I or the IGF-I plus zinc-induced increases in cellular protein, although okadaic acid entirely blocked the IGF-I or the IGF-I plus zinc effect. The present study demonstrates that the anabolic effect of IGF-I in osteoblastic cells is enhanced by zinc ion. The enhancement by zinc may be mediated through the signaling pathway of protein kinase C and protein phosphatase in the cells.
Effect of zinc sulphate and zinc methionine on growth, plasma growth hormone concentration, growth hormone receptor and insulin-like growth factor-I gene expression in mice.

1. The current experiment was conducted to investigate the effect of zinc sulphate (ZnSO4) and zinc methionine (Zn-Met) on growth and their effect on plasma growth hormone (GH) concentration, growth hormone receptor (GHR) and insulin-like growth factor I (IGF-I) mRNA expression in mice. 2. Ninety male KunMing (KM) mice were randomly divided into three treatments. <b>The control group was fed on a basal diet containing 11.67 mg/kg of zinc. The ZnSO4 group and Zn-Met group were fed on the diets supplemented with ZnSO4 or Zn-Met at 30 mg/kg (containing zinc of 40.05 and 40.75 mg/kg, respectively).</b> The mice were offered the test diets for 10 days. Weight gains and food intake were measured at the end of the experiment, zinc contents in liver and serum were determined using atomic absorption spectrophotometry; GH was determined by radioimmunoassay, the levels of GHR and IGF-I mRNA were determined with reverse transcript polymerase chain reaction. 3. <b>Both ZnSO4 and Zn-Met enhanced weight gain and food intake in the mice, Zn-Met improved the growth and food intake more effectively than ZnSO4 did (P < 0.05).</b> The both forms of zinc had no effect on GH and the level of GHR mRNA expression (P > 0.05) and they up-regulated the expression of IGF-I mRNA (P < 0.05). As compared to ZnSO4, Zn-Met enhanced the level of IGF-I mRNA significantly (P < 0.05). 4. Both ZnSO4 and Zn-Met had no effect on plasma GH and the expression of GHR mRNA, but they enhanced the expression of IGF-I mRNA. <b>Zinc methionine enhanced the weight gain and up-regulated IGF-I mRNA expression more effectively than ZnSO4.</b>

Big'r zei:Dus zoals ik het begrijp wordt het anabole effect van IGF-1 gereguleerd door zink, maar wat zijn osteoblastic MC3T3-E1 cellen?
Er wordt hier gezegd dat dit werkt bij zinksulfaat en niet bij een andere gecheleerde zinkvorm.
Hoe zit dit dan met de zinkvormen die je in de winkels koopt? Zijn dit wel effectieve vormen?
Stefanality zei:ik heb hier een multi waar al 30 mg zinc in zit en moet deze 2x per dag nemen. Daarnaast heb ik dus ook nog zma en neem dit al weet ik veel hoelang in.
Ik heb er iig geen problemen mee, maar wat voor schadelijks zou je dan kunnen krijgen?
Stefanality zei:dus ik heb nu 60 mg zinc van de multi en nog 30 mg van de zma. in tot. 90 mg.
met wat extra koper te nemen kan ik vergiftiging tegen gaan en dus het gehalte aan koper/zinc in rode bloedcellen zullen dan niet dalen.
In de multi zit 2 mg koper in, dus zal dat wss wel genoeg zijn of niet?
Big Cat zei:Zinc sulphaat dissocieert in het lichaam in zinc en sulfaat, terwijl een chelerende peptide de zinc blijft vasthouden. Meeste producten in de handel zorgen inderdaad voor een stijging van vrije zinc niveaus. Het is trouwens telkens dat vrij zinc die in de studies de effecten geeft.
Er is nog een andere vorm van zinc, die geen vrije zinc opleverd, die hetzelfde effect heeft. Die heb ik onderzocht met het oog op transdermale preparaten.
Big Cat zei:Je vertelt nog de helft niet <b>Zinc kan IGF-I en IGF-II weg partitioneren bij IGFBP-3 en IGFBP-5 en de IGFIIR in de richting van de IGF1R.</b> Zinc is nagenoeg het meest anabole supplement dat je kan nemen. En je zult er sneller resultaat van zien dan van pakweg creatine.
Maar het hoeft daarom echt geen ZMA te zijn hoor. 90 mg per dag voor 5 dagen, en daarna 45 mg per dag zinc supplementeren kan al na enkele weken drastisch effect opleveren. Of je nu gebruikt of niet.
Big'r zei:Houdt dit effectief in dat IGF-1 meer beschikbaar komt voor het lichaam terwijl het anders geremd wordt of minder beschikbaar is?
Nog een vraagje.
Je zegt dat zink het meest anabole supplement is dat je kunt nemen. Trek je deze conclusie vooral uit wetenschappelijke studies of heb je ook al praktijkervaring ermee?
