Wat is IGF-1?

IGF-1 is een polipeptide van 70 aminozuren en staat voor Insuline-like-growth Factor. The structuur van IGF-1 komt voor 50% overeen met die van insuline.
IGF-1 is zoals in de HGH FAQ te lezen is een hormoon wat in de lever wordt geproduceerd als gevolg van HGH. Veel van de groei bevorderende gevolgen van HGH zijn toe te schrijven aan IGF-1. Het is per persoon verschillend hoeveel HGH er omgezet wordt in het lichaam van HGH naar IGF-1, maar per i.u. HGH wat geïnjecteerd word zal rond de 4-6 MG worden omgezet naar IGF-1. De grote kracht van IGF-1 zit natuurlijk in de potentie tot celdeling en het veranderen van de cel structuur. Hierdoor ontstaan er nieuwe spiervezels en nieuwe receptoren voor testosteron. Ook zal het verder de activiteit van satelliet-cellen vergroten/activeren

Gebruikers geven aan dat normale kuurtjes beter werken voor hun gevoel dan zonder IGF-1. Waarschijnlijk is dit voor een groot deel toe te schrijven aan de eigenschap van IGF-1 voedingsstoffen beter op te nemen. Het is dus een nutrientpusher onder andere. Ze ervaren vaak afname in lichaamsvet.

Het gebruik van IGF-1

De normale dosis voor het gebruik van IGF-1 ligt tussen de 60 en 120 MCG. Bij een dosis hoger dan 80 MCG is het verstandig het te verdelen over de dag in meerdere (2-3) injecties. 2 injecties zou in principe voldoende zijn per dag wegens de halfwaarde tijd van de IGF-1(6-8 uur)
Het wordt aanbevolen IGF-1 niet langer te nemen dan 4 tot 6 weken omdat daarna ook het effect afneemt, waarschijnlijk door receptor gevoeligheid vermindering en dat het lichaam een of meer typen antilichamen produceert om de overmaat van IGF-1 te compenseren en niet te vergeten de insulinegevoeligheid.

Wat gebruikers ervaren

Gebruikers ervaren tijdens IGF-1 een grote pomp tijdens het trainen. Dit met namen bij eventuele pre-workout injecties.
Verder is er voornamelijk een daling van het lichaamsvet niet ongewoon en meer hardheid over het lichaam.

Tegenwoordig gebruiken veel mensen het in de PCT (nakuur), de meeste mensen houden hun gewonnen spiermassa beter vast.
Dit komt doordat IGF-1 zoals eerder vermeld een nutriënt pusher is.
Het heeft overeenkomende eigenschappen in dat opzicht met insuline.

Werking op de testikels

Tijdens normale kuurtjes zullen de testikels krimpen, dit is tijdens kuren te voorkomen door het gebruik van HCG.
Echter dit kan IGF-1 ook! Zelfs midden in een kuur zal het testicular atrophy voorkomen.

IGF-1 bijwerkingen

Veel gebruikers van IGF-1 zijn vermoeid over de dag, dit is dan een van de negatieve bijwerkingen van IGF-1. Dit komt ook veel voor bij het gebruik van HGH in hogere doseringen.

Bij hogere doseringen (100+ MCG) ervaren sommige gebruikers van IGF-1 hoofdpijn, misselijkheid.

Een HGH-buik kan veroorzaakt worden de enorme hoge IGF-1 doseringen voor lange aaneengesloten weken.(maar dan wel extreem hoge doseringen met combinatie insuline+hgh)

VOEDING IGF-1

Nog een bijwerking van IGF-1 is dat het gelijkwaadige eigenschappen heeft als insuline dus er is een kans op daling van het bloedsuiker niveau en soms zelfs een hypoglycemia(hypo) net als bij insuline. Dit komt gelukkig zelden voor want het is een stuk minder extreem dan bij insuline gebruik. Echter zou ik wel koolhydraat inname bij het gebruik van IGF-1 in de gaten houden. Als richtlijn zou ik in de INSULINE FAQ kijken wat betreft PWO insuline voeding. Ik zou dit aanhouden als je rond de 100 MCG PWO zet. Minder mcg heb je dus ook minder dextrose en dergelijke nodig. Maar let altijd op voor een eventuele aankomende hypo.

Bewaring van IGF-1


Indien IGF-1 is gemixt met Azijnzuuroplossing kan het zelfs 2 jaar buiten de koelkast worden bewaard zonder negatief effect op de kwaliteit:

IGF-1 + INSULINE


Allereerst wil ik vermelden dat als je geen ervaring hebt met INSULINE en IGF-1 los denk er dan niet aan om deze 2 middelen gelijk te combineren!

IGF-1 en insuline hebben samen een groter effect op uw bloedsuiker spiegel. Het kan de kansen van een hypo vergroten tot wel een tienvoud!

Hoe komt dit?

IGFbp3 is een verbindingseiwit, dus ervoor zorgt dat IGF-1 lang genoeg in het lichaam is om zijn werk te doen(daarom ook IGF-1-L3). IGF-1 heeft van natuurlijk een halfwaarde van 10 min. Maar door dit toegevoegde eiwit is de halfwaarde verlengd naar 6-8 uur . Door het gebruik van insuline wordt de halfwaarde nog verder verlengt alleen dit maakt het gebruik ook weer riskanter. Insuline verhindert de afbraak van IGFBP3 waardoor IGF-1 langer actief blijft in het bloed. Dit kan oplopen tot wel 12 uur.

Insuline(normale dosering 8-10 i.u.) zou genomen moeten worden ongeveer 45 minuten voor de IGF-1 injectie.
Echter omdat IGF-1 langer actief is kan de dosering van IGF-1 omlaag.

Doordat de kans op een hypo veel groter is zorg voor voldoende koolhydraten en houdt altijd extra bij de hand voor onverwachte reacties.

Nutriënt pushers

Beiden middelen zorgen voor betere opname van de voedingsstoffen, vooral post-workout . Echter omdat insuline dat 45 min eerder wordt ingespoten dat al voor een groot deel voor zijn rekening neemt kan IGF-1 zich meer richten op herstel en eventuele celdeling.

Voeding tijdens IGF-1+INSULINE

Voor een goede guideline wat betreft de post-workout voeding refereer ik naar de algemene insuline FAQ.
Hou dit nog strenger aan en verhoog zelfs de koolhydraat inname iets. De kans op een hypo is nog groter!

Koop Insuline naalden
De makkelijkste om mee te werken zijn:
Microfine insulinespuiten 0,5ml U100 0,33mm (29G) x 12,7mm (25 euro per 100 stuks)

Er gaat dus 0.5 ML in deze 0.5 ML is verdeeld in 50 streepjes (per 10 een grotere)

IBE IGF-1 komt in 1000MCG gevriesdroogde poedervorm. Je krijgt er 1 ML AA vialtje bij. Mix deze samen en dan heb je gewoon weer 1000 MCG/ML net als van MR.
Dus voor 60 MCG heb je 0.06 nodig (6 streepjes op je insuline naald)
Dus voor 80 MCG heb je 0.08 nodig (8 streepjes op je insuline naald)

GG IGF-1

GG IGF-1 komt ook in gevriesdroogde vorm. Je krijgt bij deze 1.5 ML AA bij.
Als je hiervan maar 1 ML AA toevoegd bij het poeder heb je gewoon 1000 MCG/ML net als bij MR en IGF-1.

Kies je voor de volledige 1.5 ML (want moeilijker rekenen is en meer brand) dan heb je 1000 MCG/1.5 ML oftewel 666 MCG/ML
Dus voor 60 MCG heb je 0.09 nodig (9 streepjes op je insuline naald)
Dus voor 80 MCG heb je 0.12 nodig (12 streepjes op je insuline naald)

Maar hier zie je dus ook het niet echt handig werk, dus pak liever gewoon 1 ML AA en dan zit je weer gewoon op:
Voor 60 MCG heb je 0.06 nodig (6 streepjes op je insuline naald)
Voor 80 MCG heb je 0.08 nodig (8 streepjes op je insuline naald)

LR3 IGF-1 is een Peptide die sinds een aantal jaren gebruikt wordt in de sportwereld om prestaties te verbeteren. Sinds de komst van de LR3 versie, zijn er heel wat veteranen en deskundige gaan experimenteren om de meest efficiënte en effectieve manier van toedienen te vinden. Deze pioniers hebben de hedendaagse protocollen voor het gebruik van LR3 IGF-1 vastgesteld via trial and error, zoals IKKE heeft beschreven in de bovenstaande post.

Zoals bij a.a.s zijn nog lang niet alle werkingen op het menselijk lichaam bekend, daarnaast is het een feit dat elk persoon uniek is en dat een stof een andere uitwerking kan hebben op elk individu. Wanneer we de doseringen die men in deze tak van sport toedient in acht nemen, is het duidelijk dat deze niet in verhouding staan tot de medische voorschriften ten aanzien van de doseringen.

Bij de huidige LR3 IGF-1 toediening protocollen zien we dat de werking van het product na 3 weken sterk afneemt. Een logische verklaring wordt gezocht in verzadiging c.q degradatie van de receptor waar de IGF-1 aan hecht.

Sinds een aantal jaren is LR3 IGF-1 legaal verkrijgbaar in de VS als research peptide. Hierdoor is het gebruik van het product aanzienlijk toegenomen onder de recreatieve sporters, wat ervoor heeft gezorgd dat het product nu ook steeds vaker onderzocht wordt door researchers.

Deze researchers en veteranen komen nu tot de conclusie dat de huidige manier van LR3 IGF-1 toedienen snel zal leiden tot verzadiging c.q degradatie van de receptoren, wellicht sneller dan 3 weken. Ook geven zij aan dat de doseringen die men gebruikt veels te hoog zijn.

Zij zijn ervan overtuigd dat LR IGF-1 lokale groei kan veroorzaken, wanneer deze direct na de training intramusculair (in de spier) geïnjecteerd wordt.
Maar wanneer er te hoge doseringen worden gebruikt, zullen de IGF-1 receptoren in de spier snel bezet en verzadigd raken. De LR3 versie heeft een lange halfwaarde tijd van wel 6 tot 12 uur, exact is niet bekend, dit zal dan ook zeer waarschijnlijk per individu verschillen.
Door de lange halfwaarde tijd en het feit dat de LR3 versie niet gebonden wordt aan bindingsproteinen zoals IGFBP3, heeft de LR3 IGF-1 de kans om veel langer in het lichaam rond te “zwerven” op zoek naar verse en beschikbare receptoren. Het is bekend dat de ingewanden en organen in het menselijk lichaam veel IGF receptoren bevatten, het is daarom dan ook meer dan waarschijnlijk dat de resterende LR3 IGF-1 zich daar aan zal gaan binden. De Hyperplasia (celverdeling) die daar ook zal plaatsvinden is absoluut niet gewenst. Daarvoor opteert men nu voor kleinere doseringen per injectiemoment.

Huidig gebruik van LR3 IGF-1

Op dit moment zijn er een aantal protocollen ten aanzien van het gebruik. De meest gangbare zijn op het moment:

- Totale dosering verdelen: de helft na het opstaan en de andere helft direct
na het trainen.

- Totale dosering direct na het trainen.

(time on= 4 weken, time on= time off)



Het huidig gebruik is gebaseerd op 4 weken omdat de IGF receptoren verzadigd raken, waardoor de LR3 IGF-1 niet meer het werk kan doen waarvoor het bestemd is.

Nieuwe Methode

Echter wijzen de recente bevindingen naar een ander regime voor het gebruik van LR3 IGF-1.

Er wordt verondersteld dat exogene IGF-1 tijd nodig heeft om volledig geabsorbeerd en verwerkt te worden door de receptor, waar het vervolgens andere transscripties ondergaat om ten slotte Hyperplasia te bewerkstellen. In het voorbeeld (1) hieronder wordt grondig uitgelegd hoe de receptor werkt en hoe de desbetreffende stof, heel simpel gezegd, wordt opgenomen en verwerkt. Er wordt aangegeven dat de receptor ongeveer 24 uur nodig heeft om te herstellen van het verwerken van de (in dit geval) exogene IGF-1.
Dan kan de receptor pas weer de nieuwe dosis LR3 IGF-1 volledig benutten.

Dit betekend dat elke dosis die binnen 24 uur na de laaste IM injectie (doelend op specifieke spiercel Hyperplasia) is toegediend, niet volledig benut kan worden door de receptor en dus op zoek gaat naar “nieuwe” of beschikbare IGF receptoren  ingewanden en organen. Wat in feiten betekend dat je kostbare IGF nutteloos is geworden.

Daarom is men nu van mening dat LR3 IGF-1 het beste tot zijn recht zal komen wanneer het om de dag geinjecteerd wordt, wellicht zelfs elke 3e dag.

Doseringen

Daarnaast wordt geadviseerd om 20mcg tot 60mcg per injectiemoment te verdelen over de getrainde spieren. Hierbij wordt verondersteld dat elke dosis hierboven niet verwerkt kan worden door de IGF receptoren in de spieren, waardoor het dus op zoek zal gaan naar andere receptoren, de receptoren in de organen en ingewanden.

Deze nieuwe manier van IGF-1 toedienen geeft de mogelijkheid om het product veel langer te gebruiken en maakt het kostenplaatje voor het gebruik van LR3 IGF-1 een stuk aantrekkelijker.

Wanneer de gebruiker merkt dat de werking van IGF met dit protocol stagneert, wordt er geadviseerd om een aantal dagen te stoppen (ongeveer 5 dagen) en vervolgens over te gaan op elke 3e dag injecties.

Uiteraard is het verstandig om op een bepaald moment een tijd off in te lassen om ervoor te zorgen dat de receptoren volledig kunnen herstellen.

Op deze manier wordt het dan ook erg interessant (met name de Kosten) om met IGF-1 te bridgen tussen a.a.s cycles.

Here is a clipping from a book I have been reading called 'Molecular Cell Biology', and I happened put some of it online. I found this extremely interesting, however. Although the exact half life of LR3 IGF-1 is not known, although estimates of 8-36 hours are what people have been saying (very big range). It is hard to say, however, after reading the following passage you will begin to see the reason that LR3 IGF-1 could only be lasting 3-4 weeks in most people.

EGF is used as an example but most growth factors activate the tyrosine kinanse receptor, which involves phosphorylation to accomplish activation. This phosphorylation causes the cytoplasmic end of the receptor (the part inside the cell) to open up and allow gene transcription in the cell to take place. However, once this occurs it takes time for the receptor to "recover", therefore, the constant administration of growth factors, especially growth factors with half lives as long as LR3 IGF-1, can cause degradation of the receptor and continually do so to multiple receptors, eventually (usually after 3-4 weeks) reducing the receptor count enough to see diminished gains.

One might speculate that it would happen faster than this, however, IGF-1 also has a binding affinity (although not extremely high) for the insulin receptor, so it isn't only the IGF-1R that is affected. This "appears" to be the reason why we see diminished gains, based off of the literature I am reading. We basically need to allow these receptors to recover, which is probably around 12-24 hours for all tyrosine kinases, and it would appear that the active life or LR3 IGF-1 is roughly 24, although this isn't definite. Hence, it would also appear that the best route to take would be an EOD injection or POSSIBLY E3D injection scheme.

Here is an idea of what the receptor looks like and it also shows the cytoplasmic tail end of the receptor and how it works.

Receptors for Many Peptide Hormones Are Down-Regulated by Endocytosis

The principal mechanism for down-regulating the receptors for many peptide hormones (e.g., insulin, glucagon, EGF, and PDGF) is ligand-dependent receptor-mediated endocytosis. In the absence of EGF ligand, for instance, the EGF receptor is internalized with bulk membrane flow. Binding to EGF induces a conformational change in the cytoplasmic tail of the receptor. This exposes a sorting motif that facilitates receptor recruitment into coated pits and subsequent internalization. After the receptor-hormone complex is internalized, the hormone is degraded in lysosomes a fate similar to that of other endocytosed proteins, such as low-density lipoproteins (see Figure 17-64). Unlike the low-density lipoprotein (LDL) receptor, internalized receptors for many peptide hormones do not recycle efficiently to the cell surface.



In the presence of EGF, for instance, the average half-life of an EGF receptor on a fibroblast cell is about 30 minutes; during its lifetime, each receptor mediates the binding, internalization, and degradation of only two EGF molecules. Each time an EGF receptor is internalized with bound EGF, it has a high probability (about 50 percent) of being degraded in an endosome or lysosome. Exposure of a fibroblast cell to high levels of EGF for 1 hour induces several rounds of endocytosis, resulting in degradation of most receptor molecules. If the concentration of extracellular EGF is then reduced, the number of EGF receptors on the cell surface recovers, but only after 12 24 hours. Synthesis of new receptors is needed to replace those degraded by endocytosis, which is a slow process that may take more than a day.

The fewer hormone receptors present on the surface of a cell, the less sensitive the cell is to the hormone; as a consequence, a higher hormone concentration is necessary to induce the usual physiological response. A simple numerical example illustrates this important point. Suppose a cell has 10,000 insulin receptors on its surface with a KD of 108 M. As noted earlier, in many cases only a fraction of the available receptors must bind ligand to induce the maximal physiological response (see Figure 20-7). If we assume only 1000 receptors must bind insulin to induce a physiological response (e.g., activation of glucose transport), we can calculate the insulin concentration [H] needed to induce this response from Equation (20-2) rewritten in the following form:

where RT = 10,000 (the total number of insulin receptors), KD = 108 M, and [RH] = 1000 (the number of insulinoccupied receptors). In this example, the necessary insulin concentrations is 1.1×109 M. If RT is reduced to 2000/cell, then a ninefold higher insulin concentration (108 M) is required to occupy 1000 receptors and induce the physiological response. If RT is further reduced to 1200/cell, an insulin concentration of 5 × 108 M, a 50-fold increase, is necessary to generate a response.

Experiments with mutant cell lines demonstrate that internalization of receptor tyrosine kinases plays an important role in regulating cellular responses to EGF and other growth factors. For instance, a mutation in the EGF receptor that makes it resistant to ligand-induced endocytosis or, in dynamin, that blocks formation of clathrin-coated endocytic vesicles substantially increases the sensitivity of cells to EGF as a mitogenic signal. Such mutant cells are prone to EGF- induced cell transformation. Interestingly, the mutant-dynamin inhibition of internalization also causes a qualitatively different pattern of phosphorylation of substrate proteins by the activated EGF receptor, as well as quantitative changes in the phosphorylation of known components in the EGF signaling pathways. Interestingly, internalized receptors can continue to signal from intracellular compartments prior to their degradation. This raises the intriguing possibility that receptor activity can be spatially controlled. Hence, internalization may modulate both the nature of RTK-transmitted signals, their magnitude, and location.

Studies with RTKs that bind PDGF suggest that PI-3 kinase plays an important role in the endocytosis and down-regulation of this class of receptors. Mutations that abolish the ability of the PDGF receptor to bind PI-3 kinase but not other enzymes (e.g., PLCg) cause a reduction in the rate of receptor degradation. Although the mutant receptor is internalized, its sorting to the lysosome for degradation is blocked by an unknown mechanism. The observation that yeast cells expressing a mutant PI-3 kinase exhibit defective sorting of proteins to the vacuole (the yeast lysosome) raises the intriguing possibility that this enzyme plays an important role in membrane trafficking both in yeasts and mammalian cells.

On July 20 I got into some pretty intense discussion on another board about IGF-1. I got so rattled with the misinformation that I decided to loose my 13 years of reading on IGF-1 onto that board. Here's the result.

This is false.

The difference between rhIGF-1 and Long R3 is that the Long R3 does not get bound by binding protein and thus is 100% active whereas you do lose a great % of whatever amount of rhIGF-1 you inject to IGFBP3.

While technically it is true that if you inject a large amount of the rhIGF-1 it will have almost only localized effect, it is so because the "excess" that does not bind to cells in the muscle in which it is injected is rapidly bound up by IGFBP3 and thus rendered unusable by cells elsewhere. It would be much much better in such a case to inject a smaller amount and not have ANY excess that gets bound up by IGFBP's.

And while technically it is true that if you inject a large amount of Long R3 IGF-1 in a muscle, it will first bind to the nearest available receptor, and spread, binding to more and more receptors and not be bound up and neutralized by IGFBP's, meaning that it will travel all through your body and grow all kinds of tissue. This is called the systemic effect of IGF-1. Therein lies the only distinction in terms of BOTH half-life and localized/systemic effect between the Long and the human varieties.

What does all this mean?

It means that technically, for the part of the muscle in which you inject, THERE IS NO DIFFERENCE BETWEEN rhIGF-1 and Long R3 IGF-1. They both have the EXACT SAME LOCAL EFFECT. But rhIGF-1 gets neutralized quick, whereas Long R3 gets to float around until it finds a receptor.

What does all this tell us?

It tells us many things. Let's start with what we want, then see where that leads us. What do we want? Bigger muscles. More muscle cells that we will later grow with exercise and gear. A pump? Fatloss? Yeah, right. You can get a pump with a good "pump" product for a quarter of the price of IGF-1. Fatloss? clenbuterol/Alb and T3/T4 will give it to you again at a fraction of the price of IGF-1. More muscle cells, you can ONLY get with IGF-1 (and MGF too). Nothing else will give it to you and if you are using IGF-1 for anything else, you are misusing it. More muscle cells is CLEARLY the best use for IGF-1.

What does all this tell us?
It tells us that we should use IGF-1 to make more muscle cells. It's the only thing that can give it to us and more cells is more growth, which is our goal.

What does this tell us?

The localized effects are the best. Long R3 IGF-1 can float around your body and attach to anything that has IGF-1 receptors. The intestines is the place that has the MOST IGF-1 receptors and it also happens to have lots of blood flow. Injecting large amounts of Long R3 ENSURES that you are growing your intestines. Remember, more cells doesn't equal more size right away. Wait a bit, and see them grow.

What does this mean?

It means that if you are injecting upwards of 50mcg of IGF-1 you are growing your intestines. Yes you are also growing muscle and you may be getting leaner in the process. Your waistline looks trimmer. Nice. A few months down the line, your new intestinal cells will be of their full adult size and you will have acquired the perma-bloat look. Guaranteed. Maybe not Coleman-size perma-gut, but SOME perma-gut and it will keep growing. Guaranteed. Just as your new muscle cells can keep growing and growing IF you pin IGF-1 in a way to maximize new muscle cell creation.

HOW?

Heavy resistance exercise strongly upregulates the IGF-1 receptors on the stressed muscle. That means that after your workout, the muscles you trained are at their BEST STATE for receiving IGF-1 and growing many new cells. That's when you pin. This upregulation of IGF-1 receptor during exercise is short-lived. The science is not readily available so I am unable to quote a paper, but within 60 minutes of the last set, the receptors are back at baseline. This means, PIN IMMEDIATELY POSTWORKOUT and you will get your new muscle cells. PIN A LESSER AMOUNT and you will get only new MUSCLE cells out of your IGF-1. Pin more and you will grow other things, including stuff you wish you didn't grow.

What else?

All the talk about IGF-1's half-life is UTTER BULLSHIT. It is technicality without any real-world applicability. Yes rhIGF-1 has a "short half-life". But what does it mean? It means that it is either taken up by a cell receptor or bound up by a binding protein in short order. Does it mean that 20 minutes after the IGF-1 is pinned you should pin more because "blood levels are low"? Not by any means. Once it's activated a cell receptor, that's where it initiates a cellular response that will take about 72 hours to be complete and which will consume lots of energy. So the half-life of 20 minutes means NOTHING BECAUSE THE EFFECTS STILL LAST 72 HOURS ALL THE SAME.

What about Long R3 IGF-1?

Yes technically it has a longer half-life. Why? Because it either gets rapidly taken up by a cell receptor or... Just floats around. Until it can find a receptor or is destroyed by the immune system or some other metabolizing mechanism. BUT THIS MEANS ***NOTHING***!!! Why does it mean nothing? BECAUSE once it attaches to a cell receptor, it initiates a cellular response that will take about 72 hours to be complete. THIS CELLULAR RESPONSE IS ALL THAT INTERESTS US. Not "blood levels", that's utter bullshit. As a matter of fact, the one thing YOU DO NOT WANT IS FOR BLOOD LEVELS OF IGF-1 TO BE ELEVATED. Because that means you are growing everywhere and this means first and foremost your guts. Sure it feels like it's working while you're on. Just you wait 9 months and see that you look like Craig Kovacs. Bravo, you now have the biggest intestines in the world.

Half-life means nothing. Localized vs systemic = bad argument. You want localized effects. Period. You get them by pinning immediately postworkout. Period. End of argument.

OMFG I am so tired of all the misinformation floating around on IGF-1. Look at the length of this post. Did you read all of it? You should, you know.

40mcg is plenty. We have to realize that this is a huge amount compared to what the body naturally produces. Maybe we can ask TheGame46 who is working on his master's degree in endocrinology what the actual amount produced by a normal human, say even with exercise, but it's probably something less than 1mcg.

20mcg each side. 30 each side in the quads. That's plenty. Now, you won't see major, immediate LBM increases, but THAT IS NOT WHAT IGF-1 IS FOR. That's what AAS are for. 40-50mcg total will let you get plenty of hyperplasia, not grow your intestines too much, and save you plenty of $. The newly added muscle cells will take months to grow, but they will, and you will use IGF-1 again because it gets reasonably inexpensive with such a protocol.

Another thing: much of the newer research shows that EOD and even E3D igf-1 treatment is better than ED because ED downregulates the receptors too quick. It takes some time for receptors to be able to come back in full after a megadose of even 20mcg of IGF-1. So you may want to think about switching to EOD lifting and IGF-1 immediately postworkout every workout, or 2on/1off and pin the lagging muscle E3D. These dosing patterns won't give you pounds of immediate muscle, but they will give you hyperplasia, which means continued growth at very decent rates, and the ability to continue treatment for a long while until response diminishes.

And no Coleman guts.

Great questions. I'll start with some background on the peptides from back before IGF-1 was commonly used. GH was the first peptide to be used in Bodybuilding. We pretty much know what GH does and doesn't do and all that, so I'll skip this part. Then came along insulin. It quickly became apparent that slin on its own doesn't do much for muscle. It does make you fat but not much bigger. With AAS and tons of food, it's better. Later it became extremely clear that Slin & GH was the winner combo, the most synergistic combination around.

What few people realize even today - and it's been what, nearly 20 years of insulin usage in bodybuilding, is that the very reason why slin and GH are synergystic is that when levels of both are high, the liver turns the GH into IGF-1. That's right, when doing slin & GH, you are in fact using these because your body makes more IGF-1 with them. So it isn't the slin OR the GH nor actually the compounding of the effects of each, but rather good old IGF-1. Even the name Insulinlike Growth Factor, has been made such because of the origin of the compound in Insulin and Growth Hormone.

Now, the IGF-1 from slin & GH is not long R3 IGF-1, it's hIGF-1. It's different and possibly the effects are somewhat different than when using Long R3, especially with regards to IGF-1 receptor downregulation, which is likely much lesser with the liver-synthesized IGF-1 than with the Long R3. No studies proving this, it is theory at this point and such a study will possibly never be made, for many good reasons. One reason why receptor downregulation is lesser with hIGF-1 is its half-life, or its very limited ability to run around the body and saturate all receptors everywhere. And here we join up with the EOD and E3D protocols which state that letting the receptors rest is extremely important to continued results. You get the same effect out of slin & gh because of IGFBP3 that mops up the IGF-1 within minutes of synthesis, which makes it impossible to saturate the receptors and lets them rest. Similar effect, completely different way of achieving it.

So slin & gh are synergistic. Then the next question: what about slin & IGF or Gh & IGF? IGF is synergistic with both. MOST of the effects of GH are mediated through IGF-1 but not ALL of them. Among the good effects of GH that IGF-1 does not exert is anabolism to ligaments, for example. This is just an example to show that there is a benefit to using GH & IGF-1 at the same time. There is evidence that ED dosing of LR3 reduces GH release in the body, so it makes plenty of sense to use both at the same time.

Slin & IGF is a different animal. Most of the benefits of insulin come from its ability to increase IGF-1. Unless you are diabetic, your body makes enough insulin. Eat more, it releases more insulin. More carbs? More slin. The limit to the body's ability to release slin isn't easily reached. Even feeding 10,000 cals ED your body can produce the slin to store that. Easily.

Am I stating there is no use in pinning slin & IGF together? No. There is evidence that shows that pinning slin with IGF-1 increases the length of the effects of IGF-1. Especially the hypoglycemic effects, obviously, but this has pretty far-ranging and beneficial implications, among which saturating the lean cells with nutrients and having a low blood sugar level are not the least. Obviously they are both hypoglycemic compounds so carbs have to be adjusted up when adding IGF-1 to slin, or slin reduced. I prefer the second option, although I am at a loss as to the amount of slin you would have to remove for compensation with, say, 40mcg IGF-1.

Personally I have not done this. Both my grandfathers were diabetics, so I'm not playing with slin. Especially that I have a natural tendency to go hypoglycemic easily. IGF-1 though is simply GREAT for me.

What I did do, over 10 years ago, is use an extremely potent GH releaser named GHB and combined that with a few ounces of sugar, the idea being of course a cheap version of GH & Slin. Obviously it worked great over a few months and it did produce hyperplasia, as made very obvious by the muscle size I retained when taking a 2 year layoff from lifting because of a non-training related injury.

I have run LR3 at 20, 30, 40 and 50mcg ED as well as variations of only postworkout pinning. I suggested EOD and gapped dosing way before lab research showed that this would be a better dosing protocol.

In my experience, IMMEDIATELY-POSTWORKOUT dosing is all-important to hyperplasia. SOME benefit is had by pinning preworkout and at other times, but the vey best resutls from pinning immediately postworkout. I have experimented with 5-minutes postworkout and 20-30 minutes postworkout and have found the 5-minutes postworkout dosing to be VASTLY superior to any other dosing protocol. I know it isn't the most practical for most of us, but I'm saying what I have seen on myself.

Gains out of IGF-1 are difficult to account for. Firstly, it is much more a recomposition compound than a mass or fatloss compound. On AAS, the gains are "this many lbs of LBM". On clenbuterol/Thyroid, gains are "so many lbs of flab". On IGF-1 the gains are "some fatloss, some muscle gain/retention, and this many new cells that I will grow in the coming months".

But suffice it to say that my first experimentation protocol was 5 minutes postworkout in my biceps, delts and chest because my previous research had indicated that the postworkout window was limited, and because those were my lagging bodypart. My biceps went from 17" to 17½" in the first 2 weeks along with some fatloss and another ½" in the 2 months afterward, my dumbbell curls going from 55 x 10 to 65 x 10. That was after 12 years of natural training, with genetic potential pretty maxed out. Chest and delt results I did not even attempt to quantify but the difference was clearly visible.

On another board there was a log where the guy was shooting only his biceps because he read that local effects were little and his bis were lagging. A couple months after his log was done I asked about his biceps and he said they had now taken the lead in his muscular development.

Could this change with large doses of AAS? I could be wrong. Completely so, but with verry large amounts of certain anabolics your IGF raises drasticaly. Why would this not result in some for of perma gut?
I beleive GH can cause some organ growth correct? Maybe that has a different mechanism but it seams this only happens at verry high doses over verry long periods of time.
Why do we not see such organ growth with the use of extreme amounts of AAS over periods of years? And a more importantly, if you were to take large doses of AAS especialy those of the stronger breed do you think that the doses could increase, perhaps from increasing the rate of the receptors processing the IGF-1r3.

Also wouldn't it be verry usefull to use this chemical post cardio because of the blood pumping so drasticaly to the muscle sites, even pre or mid cardio work out?

Those are actually some very good questions. The answers are equally good.

There are two completely different ways in which IGF-1 is produced in the body. Even the IGF-1 molecule itself is slightly different in each case. The first, well known case, is where GH & Slin are used by the liver to make IGF-1 which is then released into the bloodstream. AAS has little to no bearing on this systemic, or "paracrine" IGF-1. It just circulates in the bloodstream and eventually finds an IGF-1 receptor on the outside surface of a cell and attaches to it, activating it.

The other pathway, the one that is rarely discussed, is the autocrine pathway. This is where a cell will produce its own IGF-1, a slightly different peptide than the systemic, for its own internal use. It is produced inside the cell, and acts on receptors within the cell. This is the pathway that AAS will greatly upregulate. This IGF-1 never leaves the cell.

So on one hand you have the systemic with its effects on the surface receptor and you have the autocrine with its effects on the internal receptor. So obviously when you know this it becomes obvious that the IGF-1 from AAS - the autocrine - will never give you the GH gut because the IGF-1 that it makes your cells produce never leaves the cell itself, it doesn't circulate around to go attach to an intestinal wall receptor.

The pathway through which GH causes organ growth *IS* systemic IGF-1. Most of the effects of GH are actually effects of IGF-1. GH is simply not very active on many cells but it is much converted by the liver into IGF-1 and this is what mediates the effects of GH. As I posted above, there ARE some effects of GH that are not mediated through IGF-1 but most of them are.

As far as upregulating the surface receptors through AAS usage, I have seen no evidence that points that way, but that is not entirely impossible. Improbable, but not impossible.

As far as pinning post-cardio, I don't see it. In my opinion, for bodybuilders IGF-1 has two main purposes: firstly hyperplasia, its main use, and secondly general tissue repair, meaning healing and preventing injury. Ligaments aren't repaired by IGF-1 but they're a rare exception. It is too expensive and too good at better things IMO to be wasted on a simple pump.

Indeed my friend. It should be noted that the new cells are myoblasts, pre-muscle cells, that will fuse with your existing muscle cells and donate their nucleii which are in fact myonucleii.

A muscle's protein-repair engine is the myonucleii. The more of them in a cell, the bigger the cell and the greater the ability to regenerate protein. This explains the permanent gains from IGF-1 in that the number of myonucleii does not easily decrease, which gives a cell a new minimum size. Unless of course a person undergoes starvation, but that's not the case around these parts. When we take AAS, it's the myonucleii that get stimulated into overdrive.

Indien IGF-1 is gemixt met Azijnzuuroplossing kan het zelfs 2 jaar buiten de koelkast worden bewaard zonder negatief effect op de kwaliteit:

jah maar men engels is niet echt goed begrijp er bijna niets van :s
 
Storage condition is de temperatuur waarop jij het bewaard, bij een koelkast is dat ongeveer 7C.
Je kan het op deze temperatuur ongeveer 2 jaar bewaren zonder dat er veranderingen optreden.
Let wel op, hier wordt gebruik gemaakt van AA en niet van BW.
Op andere sites heb ik ook gelezen dat als je het in de koelkast bewaard, het tot een jaar 99% van z'n werking behoud. Verder staat stond er niks over vermeld bij dat topic.

Hoelang het met BW bewaard kan blijven staat er niet, maar de meeste labs leveren IGF met AA en heb je daar dus geen last van.

Ga er dus vanuit dat je het 1-2 jaar in de koelkast kan bewaren. Maar waarschijnljik is het daarvoor al wel op :).

Gemixt met BW is het maar 48 uur stabiel:

GenSci's IGF is reconstituted using bacteriostatic water because they have secured the correct pH level required so as to not use complicated methods of reconstitution. This is beneficial for various reasons beyond the comprehension of the not-so-chemically-minded amongst us. However, it has a life of only 48 hours thus using the whole 100mcg bottle within that time frame.

Ik heb gelezen dat subq en IM in de buik injecteren eerder leid tot groei van de ingewanden. Is daar echt sprake van als je 50 mcg zet? Of moet je dan tot echt hoger zitten?
Ik weet dat iedereen anders reageert op igf in deze context, maar het leek mij nogal sterk omdat er genoeg spierreceptoren zijn voor 50 mcg eod..
En maakt het dan ook echt uit of je het in je buik zet of ergens ander, en maakt subq en IM veel verschil??

Op deze vragen kon ik geen antwoord vinden.. thnx iig ;)

Een paar tips:
Ga studeren over igf.
Wacht nog een paar jaar met juicen.

Dit werkt namelijk heel anders dan aas, maar daar kom je wel achter als je er in verdiept.

je injecteert igf nooit im in je buik!
je kunt onderhuids zetten maar...het idee achter im zetten direct na t trainen is dat je lijf veel gevoeliger is voor de igf, de igf beter wordt opgenomen en zo niet de kans krijgt, of beduidend minder, om rond te gaan reizen door t lijf en daar bv te gaan hechten aan je darmwand

Is dat ook de reden dat als je 10 minuten na training in de getrainde spier spuit, je plaatselijke groei krijgt? Want dat lees ik ook wel vaak igf-1..
Dit is echt zo'n stof waar vanalles over gezegd wordt, en dan wordt het zo niet geloofwaardig meer haha :D K*tspul.. :P
